Prenatal exposure to polychlorinated biphenyls (PCBs) can adversely affect thyroid function in the adult. A number of epidemiological studies have shown a relationship between PCB exposure and thyroid function in the offspring; however, the mechanisms for the effect of PCBs on the developing thyroid remain unclear. Our findings in the adult rat suggest that the prenatal exposure to PCBs may alter the activity of a number of thyroid regulatory peptides, including the growth hormone-releasing hormone (GH-RH) and the thyroid-stimulating hormone (TSH), thus explaining the PCB-induced hypothyroidism. However, this is a very complex mechanism since the effect of PCBs is sex specific and depends on the dose and duration of the exposure. To further explore these mechanisms we have investigated the activity of GH-RH, TSH, and thyrotropin-releasing hormone (TRH) in the fetal brain of rat exposed to different doses of PCB 153 (40, 200, and 800 ppm) by gavage from embryonic day 12 to embryonic day 16. The animals were decapitated at birth (day 0) and blood samples were collected. The brain was removed from the skull and the hypothalamus and anterior pituitary were dissected. GH-RH, TSH, and TRH were measured in the pituitary and hypothalamus. The results show that exposure to a high dose of PCB (800 ppm) caused a significant decrease in the GH-RH content in the hypothalamus; however, no effects were observed in the pituitary. The high dose of PCB 153 also caused a decrease in the TSH content in the pituitary, but not in the hypothalamus. The results for the TSH content in the pituitary are consistent with our previous findings in the adult rat where exposure to PCBs causes a decrease in the TSH content. The decrease in the hypothalamic GH-RH content observed at day 0 in the 800 ppm group cannot be explained by changes in the pituitary content of this hormone. These results suggest that the decrease in the GH-RH content in the hypothalamus may cause a decrease in the TSH content in the pituitary, which in turn may cause the decrease in the thyroidal TSH content observed in the
